Factors influencing DNA extraction from human skeletal remains: bone characteristic and total demineralization process

Abstract Background The extraction of DNA from skeletal remains with good quality and quantity is often challenging for the ability to generate typing. Previous studies demonstrated total demineralization fresh teeth bones; however, application in old has been less performed. To obtain high yield amount extracted remains, objective this study was focused on exploring factors influencing process developing effective methods. Results concentration EDTA found significantly enhance calcium chelation bone while pH solution, incubation temperature, time, volume solution were not significant. optimal condition obtained Placket-Burmann results represented good-quality highest yield. Subsequently, STR typing some specimens processed by prior improved. Conclusions a key factor chelate human remains. probably improved profiles.
